Can a Christian lose his salvation? How do you know?

No. When we believe, we 'stand' - the perfect tense of a stand taken once, with the result that we stand forever. Our salvation rests not on our grip on God but on Christ's finished work; Paul says we are saved now, and will be saved tomorrow, next year, and forever.
